MTA

Cell lines must be transfered under MTA (Material Transfer Agreement). Users must enter MTA with Tottori Bioscience Foundation through POC Inc. (agent) before order and obtain research license.

Order and payment

Order(s) for cell line should be placed after execution of MTA. Pre-payment is necessary. Cost for initial transfer includes research license for two years.

Shipment

POC Inc. will send cell lines and other products ordered by users. Users' accounts will be used for shipment. Users can use cell lines following standard protocols.

How to renew license

Maintenance (Renew)

You need to renew MTA and pay license fee every two year. You can change your research license to other types of license e.g. CRO license.

Additional supply

You can obtain new vial of cell line(s) and/or reagent from us anytime you need.

Termination

Users can terminate MTA anytime. Paid costs including license fees should not returned. Before effective date of termination users must return all cell lines and derivatives of cell lines to Torroti BIoscience Foundation.

Assays with validated protocols

We are distrbuting cell lines for validated protocols for skin sensitization assay (THP-G8) and immunogenicity assay (2H4). 

IL-8 Luc assay: Skin sensitization assay

THP-G8 cells are utilized for the in vitro skin sensitisation test (IL-8 Luc Assay), which is approved as an internationally recognized testing protocol by the Organization for Economic Co-operation and Development (OECD).
The THP-G8 cells are derived from human acute monocytic leukemia cell line (THP-1 cells) stably introducing luciferase genes, in which orange-emitting luciferase (Stable Luciferase Orange, SLO) is expressed under the control of interleukin-8 (IL-8) promoter, and red-emitting luciferase (Stable Luciferase Red, SLR) is expressed under the control of GAPDH promoter, used as an internal control reporter.
To evaluate skin sensitisation, transcriptional activation of interleukin-8 (IL-8) promoter by test chemicals, seen in human dendritic cells in skin, is simply measured by luciferase reporter assay.
*THP-1 cells were established by Department of Pediatrics, Tohoku University School of Medicine
* THP-G8 cells were established by Department of Dermatology, Tohoku University School of Medicine (Takahashi T. et al. Toxicol. Sci., 124, 359-369, 2011).

IL-2 Luc assay: Immunogenicity assay

2H4 cells are utilized for the in vitro immunotoxicity test (IL-2 Luc Assay), which is approved as an internationally recognized testing protocol by the Organization for Economic Co-operation and Development (OECD).
2H4 cells are derived from human acute T lymphoblastic cell line (Jurkat cells) stably introducing luciferase genes, in which green-emitting luciferase (Stable Luciferase Green, SLG) is expressed under the control of interleukin-2 (IL-2) promoter, orange-emitting luciferase (Stable Luciferase Orange, SLO) is expressed under the control of interferon (IFN)- γ promoter, and red-emitting luciferase (Stable Luciferase Red, SLR) is expressed under the control of GAPDH promoter, used as an internal control reporter.
To evaluate immunotoxicity, transcriptional change of IL-2 promoter of human T cells by immunotoxic chemicals is simply measured by luciferase reporter assay.

THP-G8 cell line

THP-G8, that harbors the SLO and SLR luciferase genes under the control of the IL-8 and GAPDH promoters, respectively, was established by the Dept. of Dermatology, Tohoku University School of Medicine. (Takahashi T. et al. An in vitro test to screen skin sensitizers using a stable THP-1-derived IL-8 reporter cell line, THP-G8. Toxicol Sci, 124(2), 359-369, 2011) (International patent publication No. WO2012/002507A1)

2H4 cell line

A Jurkat-derived IL-2 and IFN-γ reporter cell line, 2H4 cells, that harbors the SLG, SLO and SLR luciferase genes under the control of the IL-2, IFN-γ and GAPDH promoters, respectively, was established by Tsuruga Institute of Biotechnology, TOYOBO Co. Ltd. (Saito R. et al. Nickel differentially regulates NFAT and NF-κB activation in T cell signaling Toxicology and Applied Pharmacology, 254, 245–255, 2011)

TriplucTM reagent

TriplucTM Luciferase Assay Reagent is optimized for homogeneous assay of the three colors-emitting beetle luciferases expressed in mammalian cells. For IL-2 and IL-8 reporter assays users must use this reagent for best results.

Improvement of stability

To improve stability of gene cell lines containing human artificial chromosome (HAC) are under development at Tottori Bioscience Foundation.

Assay reagent: TriplucTM

TriplucTM reporter assay is a powerful tool for simultaneously monitoring the expression of three genes, based on green-emitting luciferase (Stable Luciferase Green (SLG), maximum wavelength 550 nm), orange-emitting luciferase (Stable Luciferase Orange (SLO), 580 nm) and red-emitting luciferase (Stable Luciferase Red (SLR), 630 nm). TriplucTM Luciferase Assay Reagent is optimized for homogeneous assay of the three colors-emitting beetle luciferases expressed in mammalian cells.

Luminometer

Phelios(AB-2350) is a microplate luminometer to measure multi samples with high efficiency of light detection. Phelios is available to measure the diverse wavelength of bioluminescence from 350 to 670nm in 96,384 well microplate. It achieves high sensitivity with added and extremely desired optical system for efficiency of bioluminescent detection in low noise photon counting. Besides, it accomplishes high reproducible measurement by internal temperature controller and accurate pump in repeated injection. Moreover, even in the case of different multi-colors generation, it is available to measure the samples respectively by dedicated automatic filtering function.

Basic conditions

  • RECIPIENT shall assume sole responsibility for any claims or liabilities that may arise as a result of RECIPIENT’s use of the Materials
  • RECIPIENT agrees to indemnify and hold harmless PROVIDER of and from any and all claims, demands, losses, causes of action, damage, lawsuits, judgments, including attorneys’ fees and costs, arising out of or relating to the work of RECIPIENT with Materials or products derived
  • The Materials shall remain the property of PROVIDER
  • The Materials shall be returned or destroyed no later than the expiration
  • RECIPIENT is solely responsible for compliance with all foreign and domestic laws, regulations and guidelines, etc. applicable to use of the Materials
  • Pre-payment is required

This license is included in the fee for initial transfer fee and maintenance fee

CRO/CMO license

If you want to use cell line(s) for CRO and/or CMO business you need CRO/CMO license in addition to internal research license.

Sponsored research license

If you want to use cell line(s) for sponsored reseach you need Sponsored research license.

Engineering license

For engineering of cell line(s) you need special license. To make and utilize compounds/components derived from cell line(s) you also need special license.

Different fees will be applied depending on category of user’s organization. For academic organizations and non-profit organizations academic discount will be applied and fees for these organizations are lower than for-profit organizations.

Relationship between Biospecimen Laboratories Inc. and Tottori Bioscience Promotion Foundation.
Tottori Bioscience Promotion Foundation is the developper and distributor of THP-G8 and 2H4 cell lines.

On February 24, 2024, Tottori Bioscience Pomotion Foundation and Biospecimen Laboratories, Inc. entered an agreement and Tottori Bioscience Promotion Foundation outsources marketing and part of sales activities of THP-G4 and 2H4 cell lines to Biospecimen Laboratories, Inc. Marketing and sales activities including customer support and handling & shipment, invoicing and payment collection will be done by Biospecimen Laboratories, Inc. on behalf of Tottori Bioscience Pomotion Foundation.
